1.1 Instruments and Reagents

4,6-Dimethylpyrimidin-2-Ylamine Main reagents, Guanidine nitrate, acetylacetone,

1.2 Synthetic Route

The chemical reaction equation for the synthesis of 2-amino-4,6-dimethylpyrimidine is

as follows:

1.3 Synthesis Method

Add 50 mL of water into a 150-mL three-necked flask. Accurately weigh 6.1 g of guanidine

nitrate (0.05 mol), as well as acetylacetone and anhydrous sodium carbonate in a certain

molar ratio, and add them into the three-necked flask. Stir the mixture and slowly heat it
up to a predetermined temperature between 85 - 100 °C.

After maintaining the temperature and refluxing for the set time, cool the mixture to room

temperature. Let it stand for about 20 min and then filter. Add 25 wt% sodium chloride

aqueous solution to the filter cake and pulp it for 30 min (control the temperature with

an ice-water bath, not higher than 10 °C).

Filter again. Add an appropriate amount of water to the filter cake, stir and heat to dissolve

4,6-Dimethylpyrimidin-2-Ylamine. Then add about 2 g of activated carbon, continue to

heat and stir for about 20 min. Filter while hot. Pour the filtrate into a beaker, add an

appropriate amount of solid sodium chloride,stir for 15 min, then cool it to below 15 °C

in an ice-water bath and allow crystallization for 30 min. Filter, wash with ice water, and

dry under vacuum to obtain colorless 2-amino-4,6-dimethylpyrimidine crystals. The

measured melting point of the product is 152 - 153 °C (literature value: 151 - 153 °C).
