Effects of long-term (10-month) supplementation on neurological measures Unlike the
short-term treated mice that primarily showed improvement in mechanical sensitivity,
long-term treatment with L-serine for 10 months in C133W mice improved their performance
in the rotarod test compared with the untreated condition (P < 0.05).
Compared with the untreated C133W mice, long-term treatment with L-Serine Amino
Acid Supplement also showed a trend of improvement in mechanical sensitivity
(the mechanical sensitivity threshold was 146% of that of the wild type compared with 96%).
No significant differences were observed in the hot plate test after either short-term or
long-term treatment.
After 12 months of treatment with L-serine, there was a trend of decrease in the total levels
of deoxysphingosine (doxSA) in the tissues (liver, sciatic nerve, brain, and spinal cord) of
C133W transgenic mice compared with the untreated mice. In contrast, the doxSA levels
in the mice treated with L-alanine were generally higher than those in the untreated group
or the L-serine treated group.
This L-Serine Amino Acid Supplement difference was most pronounced in the liver tissue,
where the doxSA levels increased up to 3-fold compared with those in C133W mice and
wild-type mice (P < 0.01 and P < 0.05, respectively). A similar, though not significant, trend
was also found in the sciatic nerve, brain, and spinal cord tissues. Interestingly, after
supplementation with L-alanine, some C133W mice developed skin ulcers, which is a
common feature observed in patients with hereditary sensory and autonomic neuropathy
type 1 (HSAN1).
